The effect of the degree of hydrolysis (DH) was also evaluated for such activity. Additionally, the peptide fractions with the greatest activity were isolated by using Ultrafiltration Membranes, Ion Exchange Chromatography, and Reverse-Phase High Performance Liquid Chromatography (RP-HPLC). An increase of the degree of hydrolysis was found for these methods that correspond to an increase in the antioxidant activity; besides, the high relationship between ORAC and TEAC assays, with a correlation coefficient of 0.974, fitted a double squared model. A peptidic fraction with highest antioxidant capacity was obtained by separation processes, with values 4457.9 ± 72.5 and 1409 ± 43.41 μmolTE/g for ORAC and TEAC methods respectively, and contained peptides with sequences Q(K)Q(K)PVRDQ(K) and GAHQPSG, which were determined by MALDI-TOF-TOF and manual nESI-MS/MS.

Se analizó la capacidad antioxidante de radicales oxígeno (ORAC) y la capacidad antioxidante equivalente a trolox (TEAC) de los hidrolizados de plasma bovino obtenidos por Alcalasa® 2.4 L y el efecto del grado de hidrólisis sobre cada actividad. Adicionalmente las fracciones peptídicas con mayor actividad fueron aisladas por medio de ultrafiltración por membranas, cromatografía de intercambio iónico y cromatografía líquida de alta eficiencia (HPLC) en fase reversa. Se encontró que un incremento en el grado de hidrólisis corresponde un incremento en la actividad antioxidante, además, se encontró una alta relación entre los métodos ORAC y TEAC, con un coeficiente de correlación de 0.974, ajustado a un modelo doble cuadrado. Finalmente una fracción peptıdica con alta capacidad antioxidante fue obtenida por medio de los procesos de separación, con valores de 4457.9 ± 72.5 y 1409 ± 43.41 μmol TE/g para los métodos de ORAC and TEAC respectivamente, la cual contenía péptidos con secuencias Q(K)Q(K)PVRDQ(K) y GAHQPSG, determinadas por MALDI-TOF-TOF y nESI-MS/MS manual.Universidad de Antioquia.
