Summary:Streptomyces is a group of filamentous bacteria, best known to produce diverse clinically important antibiotics. This group has a complex life cycle regulated by 3’-5’-cyclic diguanylic acid (c-diGMP). C-di-GMP is synthesized by enzymes called diguanylate cyclases (DGCs), and there are multiple genes that encode these enzymes. However, its function in regulating the life cycle is not completely clear. The deletion of the diguanylate cyclase-encoding gene, cdgB, affects the phenotype of the model species Streptomyces venezuelae, indicating its regulatory role in the organism's life cycle. In this internship, I was part of the Gallagher lab, and we aimed to implement the Bacterial Adenylate Cyclase two-hybrid system (BACTH) to determine possible candidates for protein-protein interaction with CdgB. The samples that showed positive interactions revealed alignment with various genes. A putative interacting partner (PIP) was screened directly through BACTH system, but the initial results were inconclusive.
