description |
In this thesis, we isolated bacteria belonging to the Streptomyces genus from potato fields in Colombia; we characterized them phenotypically and genotypically and evaluated potential virulence factors involved in scab disease. Based on our initial results, we designed and evaluated a CRISPR/Cas system aimed at controlling the virulence of one of the scab-causing isolates in vitro. The CRISPR/Cas based genetic modification of the isolate focused on the edition of a specific target gene that we believed could be linked to virulence in this isolate. We explored the utilization of the CRISPR/Cas system to edit this particular gene, initially to investigate its involvement in pathogenesis, but most importantly, to test whether the CRISPR/Cas system can be used to control the virulence in scab-causing Streptomyces spp., which could lead to a new tool for scab disease management. We believe that this approach could be beneficial for the control of scab disease, and to protect human health and ecosystems from the damaging effects of commonly used pesticides. |
